    Protein Summary

    Gene SO_3770 from Shewanella oneidensis MR-1 encodes the NP_719307 amino acid sequence that belongs to PhoU family of phosphate regulatory proteins that regulates (via an unknown mechanism) the high-affinity phosphate ABC transporter system (PF01865).

    SCOP classifies 2olt in the all-alpha class, PhoU-like (super)family. According to a Dali search, 2olt is structurally similar to several  other  all-alpha helical proteins like PDB:3l39 (Z-scr=22),  PDB:1sum (Z-scr=19), PDB:1t72 (Z-scr=17). 2olt has a significant similarity score to the ribosome recycling factor (PDB structure PDB:1ek8; DALI Z-score 10.1, RMSD 3.3; 12% sequence identity within 103 superimposed residues).

    Side chains of conserved Glu58, Asp62, and Asp93 point to each other - indicating the presence of a charged molecule in the vicinity (not observed in the crystal structure). Conserved Asp173, Glu169, Leu165, and Asp210 are clustered together close to two clefts (there is an EDO ligand from the medium in one of the clefts). These residues are next to Gly216 that is very close the neighboring cluster of conserved residues (Leu18, Leu73, Pro80, Arg82, Asp84, Leu85, Ile139, Leu142, Leu145, Val215, Gly216, and Met222) located at one end of the helical bundle. Other conserved residues are located inside the core of the protein. Analysis of the crystallographic packing of 2olt using the PQS server {Henrick, 1998 #73} indicates that an hexamer is the biologically relevant form.

    The exact mechanism of how  SO_3770 regulates high-affinity phosphate ABC transporter systems is unknown. Genome vicinity analysis of SO_3770 finds a strong link (STRING score 0.983) with SO_3771, a putative phosphate transporter. Other genomic neighbors of  SO_3770 have similar phylogenetic cooccurrence and may have functions interdependent:
