    Protein Summary

    The gene SO_3770 from Shewanella oneidensis encodes a phosphate transport regulator (66% sequence identity with a homologue from Vibrio vulnificus), and a member of the PhoU family ( PF01865).  

    The protein structure, 2iiu, belongs to the class of all alpha proteins and adopts a spectrin repeat-like fold type SCOP46965.  2iiu is highly similar to PhoU homologue from Thermotoga maritima 1SUM (Dali Z-score=19). The given structure does not contain multinuclear iron cluster; however it does contain the conserved motif E(D)XXXD (EEDTDD) capable of coordinating this type of iron clusters.

    Inorganic phosphate (Pi) uptake is of fundamental importance in the cell physiology of bacteria because Pi is required as a nutrient.  Two distinct systems for the uptake of Pi have been described: the low affinity phosphate inorganic transporter and the high affinity phosphate-specific transporter (PstSCAB).  When the preferred Pi source is in excess, it is taken up by the phosphate inorganic transporter.  The PstSCAB transporter belongs to the superfamily of ATP-binding cassette transporters and is encoded by the Pst operon.  The Pst operon is part of the phosphate (PHO) regulon.  The PHO regulon is repressed by the PhoU protein under conditions of  an excess of extracellular Pi. [Ref]
