    Protein Summary

    The SSO2295 (13815598) gene from Sulfolobus solfataricus encodes the NP_343666 protein, a 4-hydroxybenzoyl-CoA thioesterase (4HBT) PF03061 EC  The protein belongs to the class of alpha and beta proteins (a+b) with thioesterase/thiol ester dehydrase-isomerase fold type (beta-alpha-beta(4); 2 layers: alpha/beta) SCOP.  4HBT forms a homotetramer with four active sites.  Each subunit of the 4HBT tetramer adopts a so-called hot-dog fold.  The enzyme catalyzes the final step in the 4-chlorobenzoate degradation pathway in which 4-chlorobenzoate is converted to 4-hydroxybenzoate in certain soil-dwelling bacteria.  Several structure of the enzyme homologues from other organisms have been reported: Pseudomonas Aeruginosa 2O5U [Ref], human 2FOX [Ref].  A number of other proteins also share the hotdog fold. These proteins have related, but distinct, catalytic activities that include metabolic roles such as thioester hydrolysis in fatty acid metabolism [Ref], and degradation of phenylacetic acid and the environmental pollutant 4-chlorobenzoate [Ref].
