    Protein Summary

    TM0119 gene from Thermotoga maritima encodes the protein NP_227935, a putative acetamidase (similar to proteins from COG2421). TM0119 shows a tandem repetition (1-140; 140-280) of the PF03069 group sequence that contains amidohydrolases of formamide (EC: and acetamide. Its neighboring gene, TM0120, is annotated as putative oxidoreductase.

    SCOP classifies 2f4l in the all beta class, acetamidase/formamidase-like (super)family. 2f4l top Dali hits are with PDB:2ii1 (Z-scr=41), and PDB:3b9t (Z-scr=34). The N-terminal domain is a jelly-roll made of two greek-key motifs and is most commonly seen in viral capsid proteins. It contains extensive insertions to the jelly-roll core. It is difficult to determine if the insert region (76-120) should be classified as a separate domain. The C-terminal is a small domain with structural similarity to the SCOP fold: probable bacterial effector-binding domain (link).

    An industrially attractive L-specific amidase belongs to the same protein family ([Ref]) indicating that TM0119 may have substrate specificity for the L-enantiomers.
